D3 upregulates intestinal calcium uptake while K2 activates osteocalcin and matrix GLA protein, which shuttle calcium into bone matrix and away from soft tissue.

At typical doses (1,000–5,000 IU D3 with 100–200 mcg K2 as MK-7), the synergy is well-documented. K2 deficiency may partly explain why high-dose D3 alone has shown mixed cardiovascular results in some trials.
